The pair of tiny molecules caused a decrease in cholesterol and fat mass

Researchers at the Scripps Research Institute in Florida have developed a pair of molecules that could lead to weight loss and one day treat disorders like obesity and diabetes.
Thomas Burris, study leader and a professor at Scripps Florida, along with a team of researchers, were able to synthesize a pair of tiny molecules that are capable of altering the biological clock in animals.
The small molecules alter the circadian rhythm and pattern of core clock gene expression in the brain’s hypothalamus. Circadian rhythms react to a 24-hour cycle of light and dark while the brain’s hypothalamus relates to the daily rhythms in mammals.
The synthetic compounds activate proteins called REV-ERBα and REV-ERBβ, which are partially responsible for the biological rhythms associated with metabolism in mammals.
The researchers administered the synthetic compounds to obese mice twice a day for 12 days. After the 12 days, researchers tested the mice and found that fat mass, hyperglycemia, and cholesterol levels improved significantly. The mice were given a diet of high fat and high sugar foods, yet still lost weight.
According to the study’s results, cholesterol was reduced by 47 percent and triglycerides in the blood decreased by 12 percent.
The compounds also played a part in controlling the mammals’ activity during night and day, which the researchers believe could be used to control insomnia and other sleep disorders. Researchers found that oxygen consumption increased by 5 percent in mice during the day and night, which means increased energy expenditure, but they found that these increases were not due to increased energy – there was a 15 percent decrease in movement during the same time that oxygen consumption increased.
“The idea behind this research is that our circadian rhythms are coupled with metabolic processes and that you can modulate them pharmacologically,” said Burris. “As it turns out, the effect of that modulation is surprisingly positive – everything has been beneficial so far.”
This study was published in Nature.

diabetes my ass. this will be used by chronic over-eaters, aka KFC/mcdonalds regulars, to reduce their weight instead of exercising and maybe not supersizing everything all the time.

McDonald's doesn't use pink slime in their burgers. They are actualy much better than their competitors no matter what fantasy land you live in. They were the first big chain to remove transfat from their french frie oil and they challenged others to do the same. They stopped using pink slime this year.

They are also pushing the swine industry to have much better conditions for their nursing pigs.

But hey, don't let the facts smack you in the face.

Actually no, McDonald's announced just 2 months ago they would be be removing "pink slime" from their "burger products".

So I'd guess you either are paid buy some marketing firm to post on any articles about 'pink slime', or you are an advocate of a terrible industry producing terrible food.

I would not consider an invasive surgery to be simpler than taking a shot twice a day. People with Diabetes are already used to taking shots already.

All this really does is create the mindset that eating mass quantities of food, of which a single portion has your daily caloric intake, will continue to be acceptable.

A sedentary life style, even if you are at a healthy weight, is not good for anyone. And no form of medication will help to alleviate that.

There is an even easier solution, get off your fat ass and walk for an hour, eat veg instead of deep fried everything, and eat less.

Before anyone flames me, I was in the category of needing to get off my fat ass, and once I did I reached my ideal weight (with involved loosing roughly 100 lbs) within a year, without any effort. So really the drug is only needed for the lazy.

Your anecdotal evidence is unfortunately completely worthless as proof for how difficult it is to lose weight. Humans are not all identical genetically, some are pre-disponitioned to gain weight more easily (and conversely, not lose it as easily).

Also, it has been shown that eating food stimulates the brain's pleasure center, and it doesn't take much of a stretch of the imagination to figure this mechanism could also be affected by genetic variation, meaning if you have a strong pleasure reaction from eating, coupled with a propensity for gaining weight, you might easily become very obese.

So TL:DR, unfounded blanket statements FTL.

Not all people are born the same, but the idea that there are many people simply "predispositioned" for weight gain is about as anecdotal as you can get. Those with actual genetic disorders leading to overwhelming urge to eat or very low metabolism are very rare, and often very dangerous (like thyroid diseases).

Do what you will, but please, stop blaming genetics.

Actually we do not know in humans how genetics affects obesity. In obese mice strains the cause is often single gene changes. In obese humans this does not appear to be the case. However obesity in humans could be due to multi-gene interactions or perhaps to rare variants in obesity genes. There are strong evolutionary arguments to think that genetics are likely to be important in obesity. Since soon we will be able to cheaply sequence the DNA of millions of humans we should be able to answer this question.

Yes we do know. It's still a matter of consuming more calories than you burn. Genetics cannot at this time make the body run on sunlight, electricity, or other forms of energy. Food. IS. It.

You're oversimplifying a very complex problem. It is true that the only way for the body to accumulate fat is by burning fewer calories than are consumed, but this isn't a simple matter of the eater being too indulgent or too inactive. If the only thing involved was what you eat and how much you move, do you really think ANYONE could stay at a stable weight for any given period of time? Do you believe you eat the same amount, to the Calorie, every month?

Even a 10 Calorie per day difference is a 1 pound per year weight gain. That doesn't sound like much, but it adds up over a lifetime, and 10 Calories per day is insignificant. If it were all a matter of input vs expenditure, our weight would fluctuate wildly.

In reality, there are a lot of factors that go in to weight gain/loss. Gut flora, genetics, hormones, what you eat, how much you eat, etc... All of these factors determine what your body does with the food that you put into it. Also, your metabolism is more effected by certain types of activities. You could spend 6 hours on a treadmill and get less benefit than you could 1 hour of interval training. And, I could spend 1 hour doing interval training and get less benefit than you could doing 1 hour of interval training.

You can't just look at a fat person and say "You eat too much and are lazy." It just isn't accurate (in all cases).

False. It is a very simple problem.

Yes most people do stay at a fairly stable weight, gaining a few pounds as they age due to lowered metabolism vs calorie consumption.

People don't actually stay at a completely stable weight, varying a few pounds every year unless they closely monitor their weight and adjust diet or activity level to compensate.

I can and do look at a fat person and think "they eat more calories than they burn". This is not a mystical situation, not even the tiniest bit complex.

EVERYONE who applies what I wrote will have control of their weight, though too often people either don't care about their weight (which is their right to choose), care less about it than eating what they want, or eat in excess due to emotional issues or environmental ones (in cold climates, an insulating fat layer is beneficial).

All the things you mention are irrelevant. No matter how much they might play a role the bottom line is no different for anyone. So long as a person is a biological entity that burns calories for energy, reducing the calorie to metabolic ratio will result in lower fat accumulation and in extension, burning fat.

Lazy? Your word not mine. I'm not the type of person who thinks someone else should do any particular thing. I'd like it if everyone were at their ideal healthy weight but I like more the freedom to live one's life as they choose.

There are many complications and side-effects of that surgical procedure (including the fact you're anesthethized and cut open, which in of itself is a risk factor), and once performed it cannot be undone in case there are problems.

Taking a pill is trivial in comparison to this. Far, far cheaper too, might one add.

It is far from simple. There are negative side effects and it does not work for everybody. Also, some are not good candidates for the procedure. I know due to my considering it myself and attending months of group meetings. What turned me off of it is them needing to put a screen in an artery to prevent the propagation of a possible blood clot. According to my cardiologist the screens can collect plaque. I've also met several people that have had the procedure and have all sorts of issues and some over eat it. It can make you toss your cookies, but some manage to still consume too many calories to lose weight. Sometimes that causes irreparable damage. If the head isn't right, it is not going to work.

One has to weigh the risks. Sometimes it becomes a matter of having to do it to save your life.

I agree! As a child of 2 diabetic parents and a diabetic older brother, I push myself physically and eat relatively healthy to make sure the chance of me developing diabetes is minimized. While I do welcome medical innovations such as this, I agree with you that (like stomach stapling), this remedy would most likely be used by over-eaters and people who want a chicken-sh^% alternative to maintaining physical fitness.

Seriously just another drug for the doctors to push. Type 2 Diabetee, and obesity are gifts from our flaundering government that legalized poisons in our food like high fructose corn syrup, and aspertame. Ever since the FDA approved the use of those chemicals in our food obesity and diabetes has been on the rise.

Paranoid much? I actually think that "they" simply don't care as long as "they" make money. I don't think that it's a huge conspiracy. Your point is well taken though. I'd invite you to look at turkeys and chickens as well. The Butterball broadbreasted white turkey is a monster that won't live much past a year because it is so massive that the poor things heart wears out to feed the enormous breast muscles (turkey cardiomyopathy!). They have no flavor aside from whatever is injected into them by the processor. Chickens are so medicated (we were taught to use tylosin powder in the water) so that they can be raised in huge numbers in tiny areas that they have osteoporosis from lack of load bearing exercise, and they have almost no flavor either. Have you had a real, family farmed chicken recently? Real chicken flavor, bright yellow color from the fats, and much less meat than you'd expect for the weight-- because their bones have normal mass to them! Education is so important. People need to know that real food doesn't look perfect, but tastes great! I also love that dairy got it's own section of the food guide. How much do you think that cost the dairy lobby? You can get EVERYTHING that's in milk from so many other sources that aren't even mentioned, even in some higher level nutrition classes!

What's your problem with that? We have evolved to survive in a world where there was an advantage to storing energy for lean periods as body fat. The world has changed. Some people have a much more difficult time than others maintaining a healthy maximum body weight. That's a lot of what science and technology are all about - making it easier or more convenient or possible to live successfully in a changed environment.
I am a bit of an excercise fanatic myself, but I am not a control freak about how others choose to live their lives. Have a little compassion, augmented by common sense.

Increase in oxygen consumption with decrease in overall activity, makes me wonder if their body temperature remained stable or increased? Oxygen metabolism is in a chemical sense "burning" a carbon based fuel, then if that energy being generated from the burning is not being converted into some type of physical action, then it should be converting into heat.

I am glad they did show a lowering of blood glucose and not just make the assumption that lowering body fat automatically lowers blood sugar, since that is not always the case. There are factors other than just body fat that block the efficient use of insulin to transport glucose into the cells, which is the main cause of high blood glucose levels. B Vitamin levels, physical activity, and other blood chemistry factors all contribute to diabetes. Also you have the fact that diabetes can cause weight gain, but the opposite is also true in that excessive weight gain can cause reduced insulin tolerance. Once you study biochemistry you learn that a living body is one of the most complex balancing acts chemically that exists and that we still know only a small amount of what actually takes place inside the body.

Their data showed both significantly lowered body fat and lower blood sugar in tandem. The two are generally linked, though as you said, not always. There are other ways to short the system.

Most likely these upregulated clocks are stimulating a large release of some hormone that's activating brown fat production and higher metabolism (the type that keeps us warm, especially as we sleep), so I would expect as you said that body temperature probably rose some. Could also be a lot of the sugar and fat was simply converted into waste products, and not directly burned.

Be very interesting to see how this develops. To think the circadian system had such a potent phenotype over fat and glucose levels is profound.

What people are starting to learn is that calories are not a number game to biological systems. Counting calories means nothing except at the extreme ends of the range. For instance, mice on a 40% calorie restriction diet actually take in more calories per pound of body weight versus mice on a normal diet--and yet they are slimmer, fitter, live significantly longer and healthier. It's a paradox.

The key is that chemical balancing act. There are so many concentration dependent and -gradients of concentrations- dependent activities and gene regulations in our bodies, it's mind boggling. But if we can figure out how to slip the body's gene expression profiles into calorie restriction mode, that would do wonders for people's health regardless of diet.

Yes it is amazing how complex the human body is. I also remember my great uncle who was about 5'7" and weighed maybe 100 pounds, walked 3 to 5 miles most days, worked on a farm most of his life, ate like a bird and still had Type II diabetes, and yet most of the commentators in the other thread know so much about biochemistry they solve the problem with "Duh, just eat less and you will be healthy". If only it was so simple.

If you want to boost metabolism there's plenty of things you can already do. Aside from obvious reasons of too much burgers and soft drinks, established DM/obesity/metabolic syndrome (where diet alone is insufficient treatment) is not about slow metabolism, its poisoned metabolism, its not about forcing what isnt working well to work faster.

It's always about the fast food, it's the butter's fault, it's people eating too much, it's a problem we are exclusive to because we don't exercise enough.

Some of you folks, and I know I'm casting a wide net here, need to grow a brain. Oops, you can't - but it's not entirely your fault.

See, the problem here is that we're all starving ourselves to death. We switched to skim milk for the school age years, we're afraid of fat, no matter the form. And God help the heathens who enjoy saturated fats (especially from animals!) and oh lord please protect us from the pure evils of coconut oil used at movie theaters!

On a more serious note however - "research" like this makes me laugh just a little. Why?

Because I was once a 430# guy who followed the advice of the Government. Why in the world do we follow the dietary advice of a government when we sure as hell don't follow them for anything else? Because everyone loves stuffing their faces, and some do to an extent more than others.

The real problem here isn't that people are eating fast food, or cooking with butter - the real problem is that we've all been a part of a grand experiment because some loose-mouthed bureaucratic blow hard conveyed his opinion stronger than the experts in a room, and then had a two bit half wit with no nutritional experience write about it to the 'merican public.

Much like the facade that fat kills, it's the knowledge we're trying to follow that is killing us.

Let me tell you, it wasn't easy going against everything I'd grown up with - especially after the last college class I took which was a nutrition class. I was preaching everything about that class - it "opened my eyes" but when in fact, it made me even more of a drone.

Fat is not the enemy. Saturated fat doesn't give you a heart attack moments after you consume it.

Eating too much of anything will make you fat and give you health problems, but cutting out fat is KILLING US.

Look at the incidences of autism, obesity and attention deficit problems. Then look at how the scale tips when you compare it to our average daily diet.

I changed my eating habits from consuming a lot of carbohydrates - upwards of 300+ grams a day, and many more times when I was at a gym following their instruction for "healthy" weight management. What did I have to show for it?

I worked my ass off for 14 weeks and I lost 5 lbs. What a joke.

I dropped the carbs, switched to fats, kept the protein and you want to know something incredible?

I lost weight.

So much so in fact, that after a month, I had already lost 8 lbs. Without exercising (got busy, my bad) so what happened when I picked up the exercise again? Fit into clothes that had been on my clothes rack for 12 years. Granted they're still large, but they fit me now.

Know what else rocks? I'm in a better mood. My attitude on life is much improved, I'm more outgoing, and I'm capable of so much more - I don't need caffeine to stay awake, I don't eat 10 times a day, and I don't even touch the crap I used to have when I had carbs in my diet still (as much as you Frito Lay people would be pained to hear it - I don't even eat chips anymore - no more nacho cheese Doritos at LAN parties... gasp!)

There's more to Obesity than what these researchers are trying to tell us. There's more to weight control than the government wants you to believe.

Why do you think they didn't do an about face when they discovered the problems were getting worse, not better?

Pharmaceuticals are making BANK. So are the universities that are encouraging this behavior. Most researchers I've met have alluded to the fact that they are pretty sure they're wrong too - that they just go where the money is. My own research and first hand experience has shown that to be the case as well.

Know what else I found out? Two weeks after I switched to cooking with butter, eating more saturated fats, and maintaining my average caloric intake of about 2300 calories (most "diet" sites say I should be at 4300 to lose weight - as if) my blood panels are awesome. My last panel was better than it was when I was in high school. This stuff WORKS. Best of all, you don't need to take anything special, you just have to change what you have been brainwashed to believe. It's a shame, I've seen so many fat folks struggle with it, opting for the surgery, and then I come and show them up with having done nothing. Figuratively - compared to them at least.

Do your own research. Find out more. Look up the paleo diet if you need a good starting point. Change your life, and don't ever let a doctor tell you that you need to take statins - those things will kill you faster than the cholesterol will.
